Sirtuins

NAD+-dependent deacylase enzymes (SIRT1-SIRT7) that couple cellular NAD+ availability to deacetylation of histones, transcription factors, and metabolic enzymes, linking energy status to chromatin and metabolism.

Sirtuins consume NAD+ to remove acetyl groups, releasing nicotinamide and O-acetyl-ADP-ribose. SIRT1 deacetylates PGC-1α, FOXO, and p53; SIRT3 regulates mitochondrial enzymes. Activation by caloric restriction, exercise, and NAD+ precursors (NR, NMN) has motivated geroscience interest. Resveratrol is a putative SIRT1 activator, though its in vivo specificity is contested.
